One of the leading theories about the origins of the HIV is that it escaped from a research lab -- probably in South Africa, since the Apartheid government was trying to engineer a virus by slicing pieces of bubonic plague DNA into it. Guess who the target victims were to be? PBS's Frontline did a piece on this about 10 years ago.

Now that we know it can be done, some asshole will probably create this and release it.

Of course, the people capable of doing this are few and far between, mostly academics... but academics tend to be the crazy ones, too. Someone with a fanatical attachment to ZPG or somebody who simply thinks the human race needs to end to save the planet... not terribly far-fetched, really, would probably not think twice about releasing this sort of thing.
