ZAZ: The Vacanti lab's technique includes what they call an "extremely important step" that was not described in the Nature paper or updated protocol published earlier: forcing the mature cells through very small pipettes for at least 25 minutes before exposing them to the acid solution.Biology workers should record video of their experiments, end to end, so they can catch such omissions. Chant "hail Mary." Use distilled water instead of tap water. Have the really strong guy tighten the lid. Whatever.Alternatively, have a lab rat undergrad do the experiment from the written protocol and see what goes wrong. I remember learning this around first grade when a teacher had me follow my own instructions, which missed a critical step. (She prompted me when I started doing something I hadn't put in the instructions because it was so obvious.)
