This is a tough one, as there's been tons of awesome characters in video games in the past 7-8 years.

But I'll try (and then see other choices and say "ohh they were awesome!"):

Favorite Playable Character:

John Marston - Red Dead Redemption.
Why: He was just a badass with a heart of gold. Simple as that. And one of the best stories out to boot.

Favorite NPC:

Garrus Vakarian - Mass Effect
Why? He was sarcastic, but would fight to the end for you and your teammates. Also one of the toughest guys in the galaxy.

Probably Sully from Uncharted. Or maybe Ezio Auditore.

I dunno, there's something about following Ezio Auditore from his adolescence to his elder years (and death, I guess) that is really great. Doesn't hurt that Brotherhood was the best game of the series (have not played Black Flag yet - waiting for a PS4 to play it on).

FemShep's got a much better voice actor. Unfortunately I played all the way through ME2 (with save transfers) before realizing that.

Luckily I found a mod tool that let you change your gender in the system file when moving to ME3.

I played through all 3 as Male Shepard.  No complaints, I actually enjoyed it.  Except the end of 3, because fark that.

She's so much better. Also when I played as boring male Shepard, I would often feel conflicted during the occasional moral dilemma. FemShep never had that issue. She is always hardcore and gets lots of scars.

My fiance played through Mass Effect (the first video she ever actually played since Nintendo days). She's one of the sweetest people I've ever known.

Saying that, she was FARKING VICIOUS in Mass Effect! She was upset that not every conversation wheel ended with a pistol whip to the face. Maybe it was FemShep subliminally urging her to do that, but she went pure renegade.

Did you play the Citadel DLC? That's definitely one of my top 5 DLCs ever. Just absolutely hilarious. All the inside jokes and Shepard making fun of his/her voice.

I played the story part of it and did little exploring, I really disliked the idea that this galaxy is at war and he wants to throw a party and hang out in the arcade. And hey everyone who could possibly hang out is here instead of doing more important things.
Just really put a bad taste in my mouth, which might be odd because again, I like the ending just fine.
I know it was a present to all the fans.....

I don't do consoles anymore, and tend to buy older games when they get cheap. Except for a few AAA titles, I generally wait for sales on Steam.

But of the more recent games I've played lately:

Playable:

Corvo Attano - Dishonored.
Catwoman - Arkham City
The Outlander - Torchlight II (No real character, but I like the gameplay style)
Zer0 - Borderlands 2 (I like sniping. And the haikus were funny.)

Non-player Characters/Villains

Wheatley - Portal 2
GladOS - Portal 2
Serana - Skyrim: Dawnguard
Tiny Tina - Borderlands 2
Handsome Jack - Borderlands 2 (Best villain dialogue since GladOS)
Mordechai - Borderlands 2 (He was my favorite character in the first one, and I thought the whole Bloodwing bit was a tear-jerker in an otherwise fairly silly storyline)

That's pretty much it. It's the ending that a lot of fans wanted, but not the one that they could put in the vanilla game. You're right, it's a huge mood shift, but it's supposed to be. While I enjoyed every second of it, the main storyline in ME3 was depressing as hell. This was a way to make it a little light-hearted again. Even in times of war, a little R&R is necessary.

I think Vaas should get his own game. There are few games where playing as the secondary villain would be amazing, and Far Cry 3 is one of them.

It's funny how so many good characters are machines. Glados as mentioned, and just a few years ago, HK-88.
