A long-term female Doctor should be someone with gravitas.  I'd..dislike, Doctor Who having a female lead based more on 'pretty'.  I think that could be a cool Doctor Who incarnation, too, but the first long term female Doctor should be older and quirky.  Just another Doctor, not really different from any of the previous except for having tits.  If that makes sense.

I've said this before and got shouted down quite a bit, but I'll say it again. Aside from off-hand comments and such, the Doctor will never be a woman. The main reason for this is because Doctor Who is primarily a kids' show and the bulk of that audience are boys. They want to play as the Doctor, play with the toys, wear the bow ties, and such. Doctor becomes a woman... now it's a girls' show and they ain't gonna watch a girls' show or play with girls' toys. Now I'm not saying this applies to all boys, but enough to affect the ratings.

Seriously, Helen Mirren.  (Or Judi Dench, or Maggie Smith)  Any of them can pull off both serious gravitas or stupid silliness when called for, and the Doctor needs both.

No. And here's why the Doctor should not be a woman; because Moffat is a bad writer. In the hands of a different, better writer I would be more up for it. But in Moffat's hand do we really want a whole series or multiple series where the Doctor's sex is the butt of jokes over and over again? Whole incredibly sexist episodes that center around her having a hard time being a woman and people around her having a hard time with her being a woman? I could go with the switch if they treated it like any other transformation, like 9 to 11 with his, "Hey, I have hair!" but instead it's, "Hey, I'm a lady. A Timelady! Fantastic!" and that was the end of it. But you know that's not going to be the case. So, please, keep it a man. The show is already suffering from bad writing. The Doctor becoming a woman and the episodes after that could be the thing that just kills the show for a few more decades.

Do the Time Lords actually change their sex randomly during regeneration?  The Doctor's got a whole history now, and randomly introducing that he can regenerate as a woman, just because people want to see a female lead, seems like it's unnecessarily complicating the story.

I wouldn't be against it as a viewer, but I think it's a bad idea storywise.

That's my feeling. If they want to have a Lady Time Lord, they can bring back Susan, Romana, the Rani, River Song, or Donna Temple-Noble.

Since NuWho, the doctors have been getting progressively younger (40s->30s->20s), so maybe it's time to get back to the roots and get someone "grandfatherly"?

The answer to that is "possibly." We've never seen it happen on screen, but the Eleventh Doctor specifically mentions one who did in a Neil Gaiman-penned episode. Most Who-Farkers seem to take that as explicit canon.

I tend to remember that the Doctor is a lying madman... that statement may have been pure nonsense.

Last season there was an episode where the Doctor mentioned one of his friends had a female regeneration. Also, while it remains controversial, Romana I was shown regenerating into what appeared to be a different species (she regenerates several times, as if trying on new outfits before deciding on Lalla Ward)
